EFFECT OF EGGSHELL SUSPENSION ADMINISTRATION AGAINST PULMONARY FIBROSIS OF WHITE RATS (RATTUS NORVEGICUS) BLEOMYCIN-INDUCED

Abstrak

Fibrosis paru adalah gangguan paru-paru yang menunjukkan pembentukan jaringan paru yang melibatkan infiltrasi sel inflamasi, proliferasi fibroblas, spesies oksigen reaktif (ROS) dan akumulasi matriks ekstraseluler yang berlebihan di parenkim paru-paru. Kebaruan dalam penelitian ini karena meneliti tentang efek pemberian suspensi cangkang telur terhadap fibrosis paru tikus putih yang diinduksi bleomisin. Penelitian ini bertujuan untuk membuktikan efek anti-fibrotik dan anti-inflamasi dari suspensi kulit telur pada model fibrosis paru tikus. Penelitian percobaan dengan rancangan post-test only control group menggunakan wirar jantan Rattus norvegicus dengan berat 200-250 g sebanyak 25 ekor tikus dibagi menjadi 5 kelompok (n = 5). Pada penelitian ini dilakukan pemeriksaan histopatologi paru-paru yang dianalisis menggunakan metode Ashcroft yang dimodifikasi dan menghitung Neutrophil Lymphocyte Ratio (NLR). Hasil penelitian menunjukkan bahwa gambaran histopatologis paru-paru pada K1 tidak menemukan fibrosis sedangkan pada kelompok K2 fibrosis paru berkembang derajat 6 hingga 7 dengan massa fibrotik besar ≥50% bidang visual. Pengurangan luas fibrosis pada kelompok P1 dan P2 menjadi derajat 4 sampai 5. Sedangkan, pada kelompok P3 derajat fibrosis adalah 2 sampai 3. Hasil ini menunjukkan bahwa area fibrosis paru menurun dengan meningkatnya dosis suspensi kulit telur. Nilai NLR untuk semua kelompok dinyatakan dalam batas normal (<4). Kesimpulan bahwa suspensi kulit telur memiliki efek anti-fibrotik dan anti-inflamasi pada model fibrosis paru-paru tikus yang disebabkan oleh bleomycin.

Abstract

Pulmonary fibrosis is a lung disorder that indicates the formation of lung tissue involving infiltration of inflammatory cells, the proliferation of fibroblasts, reactive oxygen species (ROS), and excessive accumulation of the extracellular matrix in the lung parenchyma. The novelty of this study is that it examines the effect of eggshell suspension administration on bleomycin-induced white rat pulmonary fibrosis. This study aims to prove eggshell suspension's anti-fibrotic and anti-inflammatory effects in models of rat pulmonary fibrosis. In this study, a histopathological examination of the lungs was carried out, which was analyzed using the modified Ashcroft method and calculated the Neutrophil Lymphocyte Ratio (NLR). The experimental research with a post-test-only control group design using Rattus norvegicus male wire weighing 200-250 g, as many as 25 mice divided into five groups (n = 5). The results showed that the histopathological picture of the lungs in K1 did not find fibrosis.Reduction of fibrosis area in groups P1 and P2 to degrees 4 to 5. In contrast, in the K2 group, pulmonary fibrosis developed degrees 6 to 7 with a sizeable fibrotic mass of ≥50% visual field. Meanwhile, in the P3 group, the degree of fibrosis is 2 to 3. These results show that the area of pulmonary fibrosis decreases with increasing doses of eggshell suspension. NLR values for all groups are expressed within normal limits (<4). The conclusion is that eggshell suspension has anti-fibrotic and anti-inflammatory effects on a model of rat lung fibrosis caused by bleomycin.
