Aktivitas Penghambatan α-Glucosidase Hidrolisat Kolagen dari Sisik dan Tulang Ikan Lolosi Ekor Kuning (Caesio cuning)

Abstract


This reseach aims to determine the collagen content, amino acid profile and α-Glucosidase enzyme inhibitory activity of collagen hydrolysate derived from scales and bones of yellowtail lolosi fish (Caesio cuning). Collagen extraction was performed by acid soluble collagen method using 0.5 M CH3COOH. The extraction results from fish scales and bones obtained by the ASC method obtained yields of 38 g (bb) and 12.22 g (bk). Analysis of collagen content was carried out by UV-Vis spectrophotometric method and the result was 3.63%. Antidiabetic activity test was conducted through inhibition of α-Glucosidase enzyme using microplate reader with Acarbose as positive control. The results showed that collagen hydrolysate had weak inhibitory activity with the % inhibition value at each variation of collagen hydrolysate concentration with IC50 value obtained from the highest concentration of 23.20% and did not reach IC50. Thus, the scales and bones of Caesio cuning fish have the potential as a source of collagen that is useful in the pharmaceutical and health fields.
